Abstract

Amorphophallus paeoniifolius (Dennst.) Nicolson (elephant foot yam) or commonly called suweg is one of the plants that has often been used as food and traditional medicine by the community. Suweg has various pharmacological activities and contains secondary metabolites. In addition, suweg has antioxidant, antimicrobial, anti-inflammatory, anticancer and other activities. The solution for using suweg without exploitation is by utilizing endophytic bacteria associated with suweg. Endophytic bacteria live in plant tissues and can produce secondary metabolites similar to their host but are not pathogenic to the plant. This study aimed to obtain bacterial isolates and to determine the diversity of endophytic bacteria isolated from suweg. Isolation of endophytic bacteria was carried out by maceration method on Nutrient Agar (NA) medium after surface sterilization using alcohol and sodium hypochlorite. The maceration method was carried out by serial dilutions of 10-1,10-3, and 10-5. The endophytic bacterial isolates were identified based on morphological observations, Gram staining, biochemical tests (sugar test, catalase test, motility test, citrate test, urea test, and starch hydrolysis test) and referred to several books. A total of 13 isolates of endophytic bacteria were isolated from the leaves, stems, tubers and roots of suweg. Based on morphological observations, Gram observations and biochemical tests showed that the 13 isolates of endophytic bacteria that had been isolated were closely related to the genera Azotobacter, Bacillus, Pseudomonas, and Staphylococcus. This shows that suweg has a variety of endophytic bacteria and the diversity found in leaves, stems, and tubers have the same quantity. Keywords: diversity, elephant foot yam, endophytic bacteria
