Abstract

Spathoglottis plicata Blume. is one of orchid species with lower level of genetic diversity, especially in flower color compared to other orchids. S. plicata accessions originated from Bengkulu usually produce pink to purple flower with a bright purple stems and flower stalk. Attemts to increase its genetic diversity had been done through induced mutation using Gamma irradiation with doses of 30 to 100 Gy. The genetic variation resulted from induced mutation were then identified and analysed using morphological characters and ISSR markers. Morphological characterization was conducted by applying the Desriptor and Characterization list available at BALITHI, the Ministry of Agriculture. Molecular identification was done by using ten ISSR primers available at PKBT-IPB collection. Results of cluster analysis in the second generation after irradiation treatments produced five main groups of 12 genotypes at 0.68 coefficient of similarity with the goodness of fit correlation matrix value of 0.89 (classified as suitable). The amplified product of 10 ISSR primers produced 360 bands, where 71 ISSR of loci (91.11%) was polymorphic. Principal component analysis produced five major groups at 0.68 coefficient of similarity with the goodness of fit correlation matrix value 0.91 (classified as very suitable). It was concluded that genetic diversity existed among orchid mutants and this variation would be of important for further orchid breeding programs.
