The effort to conserve and to develop indigenous banana in Bengkulu has to be done. Mostly banana in Bengkulu were infected by a fungal wilt diseases caused by Fusarium oxysporum. A tissue culture technique have an increasingly important role to play in the conservation certain plants of economic i mpor t ant , such a s bana na „A mbon Cur up‟ . Embr yo and or gan f or mat ion i nit iated f rom a mer i st em part of banana plant lead to produce high multiplication plantlet regeneration, and to improve the quality and quantity of product. In vitro organ formation could be stimulated by supplying the media with cytokinin and auxin. The results showed callus formation derived from meristem part of bana na “Ambon Cur up‟ was promoted in media with 2,4-D and kinetin. Micro shoot formation was obtained in media with kinetin. Micro shoot growth and development was enhanced by kinetin supplied in media without IBA. Supplying the media with sucrose at about 6% and activated charcoal at about 0.2% were needed during in vitro hardening. In acclimatization period, at about 100% plantlets were success transferred into acclimatization media. Plantlets growth and development were promoted in community pots filled with organic matters with Trichoderma harzianum..
